Manipulating the tropism of your delivery system via targeting systems is a promising approach to improve the precision and efficacy of lipid-based delivery systems. However, it presents new challenges including lipid and conjugation chemistry, CMC complexities, in vitro to in vivo translatability, promoting endosomal escape and avoiding accumulation in the liver.
